So I did a few searches to verify, but it looks like they never explained Chester's departure from the show. The episode ended like any other with Chester in it might end.
I think it's best that way. We can all use our imaginations to dream up possibilities of what became of Chester. Maybe he went to California with a pack on his back. But it's clear they spaced out his final appearance in a way that made the audience fairly used to his absence already. The viewership was sort of eased into Chester leaving. Festus was introduced well, Quint was still there to fill in too... it was well done.
Dennis Weaver went on to star in "McCloud" (1970-1977), a TV series where he played the title role of Sam McCloud, a deputy marshal from Taos, New Mexico, on loan to the New York City police department.
